目的研究类胰蛋白酶对血管内皮细胞(ECV304细胞系)表达干细胞因子的作用及其机制。方法采用RT—PCR方法检测类胰蛋白酶刺激后ECV304细胞炎症介质干细胞因子(stem cell factor,SCF)表达情况,用Western blot方法研究p38和JNK的表达及磷酸化。采用单因素方差分析干细胞因子mRNA的表达以及t检验分析p38和c—Jun N末端激酶(JNK)磷酸化。结果类胰蛋白酶能够上调ECV304细胞的干细胞因子mRNA的表达,类胰蛋白酶也可以诱导细胞内p38的磷酸化,以上作用均可被PAR2单克隆抗体抑制。结论在ECV304细胞中,类胰蛋白酶经蛋白酶激活受体2通过p38的磷酸化,最终上调表达干细胞因子。
Purpose To study the effects and mechanism of mast cell tryptase on endothelial cell (ECV304 cell line). Methods The tryptase-induced expression of stem cell factor in ECV 304 cells was detected by reverse transcription-polymerase chain(RT-PCR). The phosphorylation of p38 and JNK in ECV304 cells was analyzed by Western blotting. Data was analyzed by the One-way-ANOVA test and the student's test. Results Tryptase could up-regulate the expression of stem cell factor in ECV 304 cells and induce phosphorylation of p38, which could be inhibited by the anti-PAR-2 monoclonal antibody SAM11. Conclusions The results indicate that tryptase could activate p38 through PAR2 receptor and subsequently SCF expression in ECV304 cells.
