孕鼠铁缺乏对仔鼠出生健康的影响

目的探讨孕期低铁干预和成年期低铁干预对成年子鼠血脂、血糖的影响。方法将6周龄健康Wistar雌性大鼠随机分为2组,实验组喂低铁饲料(含铁7.4mg/kg),对照组喂富铁饲料(含铁320mg/kg)。与雄鼠同笼交配,低铁干预持续至哺乳期结束,随后2组仔鼠均转入富铁平衡期(10周),之后将原实验组和对照组各自再分为2组,分别给予富铁和低铁饲喂。持续16周后,检测子鼠血浆总胆固醇、甘油三酯、高密度脂蛋白、游离脂肪酸、血糖水平。结果母鼠孕期给予低铁干预组的血浆甘油三酯为1.35mmol/L、胆固醇2.0mmol/L、血糖8.31mmol/L,均显著高于对照组(P〈0.05);其肝细胞发生水样变性,胞浆淡染及疏松化,细胞体积增大,肝窦消失。结论大鼠孕期铁缺乏易致子代成年后血脂、血糖水平升高;肝脏组织细胞发生病理性改变。

英文摘要:

Objective To explore the effect and the most sensitive period of low-iron intervention in pregnant rats on serum lipids and serum glucose of its offspring in adulthood.Methods Six week-age Wistar female rats were divided into two groups randomly.The rats of experimental group were fed with low-iron diet and the rats in control group were fed with iron-sufficient diet.The rats were mated with healthy male rats after four weeks and the iron intervention were carried out untill the end of lactation period.Iron-sufficient diet were then given to all rats for ten weeks(iron balancing period).After that control group and experimental group were divided into two groups and fed with low-or sufficient-iron diet,respectively,and all the rats were killed to detect serum triglyceride(TG),cholesterol(TC),blood glucose,free fatty acids(FFA),high density lipoprotein(HDL) and leptin after sixteen weeks.Results Compared with the control group,the levels of serum cholesterol,triglyceride,glucose,weight of heart and liver of the offsprings with their mothers fed with low-level iron diet during pregnancy were elevated significantly.Besides,hydropic degeneration in hepatocytes,cytoplasm puffing and disappearance of hepatic sinusoid were observed in the rats.Conclusion Maternal iron deficiency during pregnancy will result in the increased levels of serum triglyceride,cholesteroland,blood glucose and pathologic changes of liver tissue and hepatocytes in its thirty-week-old offsprings.
