目的:研究木犀草素抑制肝纤维化进程中肝细胞上皮间质转化的作用。方法:大鼠腹腔注射50%CCl。橄榄油溶液(1ml/kg),每周2次,连续12周以复制大鼠肝纤维化模型。30只SD大鼠随机均分为正常对照(等容含5%CMC-Na溶液)组、模型(等容含5%CMC-Na溶液)组、木犀草素(100mg/kg)组。第1次腹腔注射CCI。后灌胃给药,每天1次,连续12周。HE和VG染色检测大鼠肝组织病理学变化,免疫组化法检测肝组织Ⅱ一平滑肌肌动蛋白(α-SMA)、转化生长因子(TGF)-β1、波形蛋白(Vimentin)、上皮型E.钙黏蛋白(E-cadberin)的表达。采用门静脉插管Ⅳ型胶原酶原位灌注法分离大鼠原代肝细胞。实验随机均分为空白对照(等容含10%FBS的DMEM溶液)组、模型(等容含10%FBS的DMEM溶液)组与木犀草素高、中、低浓度(40、20、10mmol/L)组。采用实时荧光定量聚合酶链反应法检测α-SMA、I型胶原(CollagenI)、Ⅲ型胶原(CollagenⅢ)、E-cadherin、转录因子Snail、白蛋白(ALB)、糖原合成酶(GS)mRNA的表达;Westernblot法检测α-SMA、Vimentin、E-cadherin蛋白的表达。结果:与模型组比较,木犀草素组大鼠肝组织α-SMA、TGF—β1、Vimentin表达减弱,E-cadherin表达增强,肝脏病理学程度减轻;木犀草素高、中、低浓度组大鼠肝组织细胞α-MA、CollagenI、CollagenⅢ、SnailmRNA表达减弱,木犀草素高、中浓度组大鼠肝组织细胞E-cadherin、ALB、GSmRNA表达增强;木犀草素高、中、低浓度组大鼠肝组织细胞α-SMA、Vimentin蛋白表达减弱,E-cadherin蛋白表达增强。结论:木犀草素具有明显抗肝纤维化的作用,其机制可能与木犀草素抑制肝细胞上皮间质转化作用有关。
OBJECTIVE: To study inhibitory effect of luteolin on epithelial-mesenchymal transition (EMT) of hepatocytes in rats with hepatic fibrosis. METHODS: Rat liver fibrosis model was induced by subcutaneous injection of 50% CCh 1 ml/kg sc twice a week for consecutive 12 weeks. Luteolin was administered intragastrieally with daily dose of 100 mg/kg once a day for con- secutive 12 weeks after the first injection of CCh. 30 SD rats were randomly divided into solvent control group (5%CMC-Na), model group (5%CMC-Na) and luteolin group (100 mg/kg). The pathological changes of liver tissue were observed by HE and VG staining. The expressions of α-SMA, TGF-β1, vimentin and E-cadherin in liver tissues were detected with immunohistochemis- try. In vitro, rat primary hepatocytes were isolated by portal vein intubation and type Ⅳ collagenase in situ perfusion method. They were divided into normal control group (blank culture solution), model group (blank culture solution) and luteolin high-dose, me- dium-dose and low-dose groups (40, 20, 10 mmol/L). The expressions of αSMA, Collagen Ⅰ , Collagen Ⅲ, E-cadherin, Snail, ALB, GS gene were detected by real-time RT-PCR. α-SMA, vimentin and E-cadherin protein were detected by western blot. RE- SULTS: Compared with model group, the expressions of α-SMA, TGF-β1 and vimentin in liver tissue were decreased in luteolin group, while the expression of E-cadherin was increased; ;pathological changes of liver tissue were relieved. Compared with model group, the expressions of α-SMA,Collagen I ,Collagen m and Snail gene in liver tissue were decreased in luteolin high-dose, me- dium-dose and low-dose groups, while the expression of E-cadherin,ALB ,GS mRNA was increased gene in luteolin high-concentrα tion, medium-concentration groups, the expressions of α-SMA, Vimentin protein in liver tissue were decreased in luteolin high-dose, medium-dose and low-dose groups, while E-cadherin protein was increased.CONCLUSIONS: Luteolin significantly at- tenuate
