目的研究人CD137配体(CD137L)在人肺癌细胞株的表达及其生物学功能。方法采用RT-PCR及流式细胞术检测人CD137L在肺癌细胞株A549、H460、A2、SK-MES-1、SPC-A1的表达水平。用细胞计数试剂盒8检测A2细胞在PBS(A组)、IgG1-FC(B组)或CD137-FC(C组)包被处理后的增殖情况,并用Annexin V/PI双染方法检测其凋亡率。结果 CD137LmRNA及蛋白在多种肺癌细胞株上均有表达。C组细胞增殖较A、B组明显增加(P〈0.05),但三组间细胞凋亡率差异均无统计学意义(P〉0.05)。结论 CD137L在各种肺癌细胞株上均有表达,CD137L介导的逆向信号能促进肺癌细胞增殖,可能在肺癌发生、进展中起重要作用。
Objective To investigate the expression of CD137 ligand(CD137L) in human lung cancer cell lines and their biological function. Methods The mRNA and protein expressions of human CD137L in lung cancer cell lines A549, H460,A2,SK-MES-1 and SPC-A1 were detected by RT-PCR and flow cytometry, respectively. A2 cells were divided into three groups of A(coated with PBS),B (coated with IgG1-FC) and C(coated with CD137-FC), and the cell proliferation and apoptosis were measured by cell counting kit-8 and Annexin V/PI, respectively. Results CD137L mRNA and protein were widely expressed in lung cancer cell lines. The cell proliferation in group C was obviously higher than that in groups of A and B(P〈0. 05), whereas there was no significant difference in cell apoptosis among groups of A,B and C(P〉0. 05). Conclusion CD137L is widely expressed in various kinds of lung cancer cell lines. CD137L-mediated reverse signal can promote cell proliferation of lung cancer and may play an important role in the development and progression of lung cancer.
