目的探讨高血压病患者血清导致血管内皮细胞结构发生变化的原因及其与高血压发生、发展的关系。方法将细胞分为高血压病组、健康组、空白组,分别用含15%高血压病患者血清、15%健康人血清、100%DMEM的培养基干预人脐静脉内皮细胞48 h,应用单细胞凝胶电泳技术(彗星实验)检测DNA损伤程度,用化学比色方法检测干预后的细胞上清中丙二醛(MDA)含量,以及超氧化物歧化酶(SOD)的活力。结果数据用SPSS 11.0软件包处理。与健康及对照组相比,用含高血压病患者血清的培养基干预后的细胞DNA损伤明显,且细胞释放的MDA升高,SOD活力降低(P<0.05)。结论高血压病患者血清中的损伤因素可导致人脐静脉内皮细胞DNA产生氧化性损伤,提示其可能与高血压病发生、发展的病理机制相关。
