目的:探讨汉族人群TLRs基因多态性与宫颈癌易感性之间的关系。方法:选取102例宫颈癌患者及100例健康查体正常人作为研究对象, PCR直接鉴定TLR2(-196 to -174 del)基因型;PCR 结合限制性片段长度多态性技术( PCR-RFLP)检测分析TLR3c.1377C/T,TLR4 Asp299Gly、Thr399Ile,TLR91486T/C及G2848A位点多态性;然后选取具有代表性的标本进行测序,对酶切结果进行验证。结果:宫颈癌组和健康对照组中均未发现TLR4 Asp299 Gly和Thr399 Ile两个位点存在单核苷酸多态性。病例组和对照组的TLR2(-196 to -174 del)、TLR3c.1377C/T、TLR91486T/C、G2848A的基因型和等位基因频率未见显著差异。结论:TLR2(-196 to -174 del ), TLR3c.1377C/T, TLR4 Asp299Gly、Thr399Ile,TLR91486T/C、G2848A多态性与宫颈癌易感性无明显关系。

英文摘要:

Objective:To study the polymorphisms of toll-like receptor ( TLR) 2 -196 to -174 del,TLR3c. 1377C/T,TLR4 Asp299Gly,Thr399Ile and TLR9 1486T/C,G2848A and susceptibility to cervical cancer in the Han population of China. Methods:A case-control study was performed which included 102 patients with cervical cancer and 100 healthy women who acted as the control group. Polymorphism at TLR2 -196 to -174 de1 was investigated by using the allele-specific polymerase chain reaction (PCR) method,while TLR3,4,9 genes were ana-lyzed by polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP ) . And the representative PCR products was sequenced to identify PCR-RFLP results. Results:TLR4 Asp299Gly and TLR4 Thr399Ile polymorphisms gene were not found in both groups. The single nucleotide polymorphisms of TLR2 ,3 ,9 have no significant differences between cervical cancer group and control group. Conclusion:TLR2 (-196 to -174 del),TLR3c. 1377C/T, TLR4 Asp299Gly,Thr399Ile,TLR9 1486T/C,G2848A gene polymorphisms may not be associ-ated with cervical cancer.
