目的 探讨补阳还五汤(BHDWD)对APP/PS1双转基因小鼠海马的保护作用.方法 雄性小鼠随机分成对照组、模型组、治疗组、BHDWD高剂量组(37.06 g·kg^-1·d^-1)、BHDWD中剂量组(18.53g·kg^-1·d^-1)、BHDWD低剂量组(9.26 g· kg^-1· d^-1).灌胃给予等体积的药物或者生理盐水,每天灌胃1次,持续28 d.采用Morris水迷宫测试行为学指标,观察小鼠的学习记忆能力,采用双抗体夹心法测定海马组织白介素-6(IL-6)蛋白水平.结果与模型组比较,BHDWD能显著改善模型小鼠的学习记忆能力(P<0.05),并且显著降低海马组织IL-6蛋白水平(P<0.05).结论BHDWD能改善模型小鼠的学习记忆功能,机制与降低海马组织IL-6蛋白水平有关.
Objective To investigate the protective effects of Buyang Huanwu Decoction(BHDWD) on hippocampal tissue in APP/PSI double transgenic mice.Methods Male mice were randomly divided into 6 groups:control group,model group,treatment group,BHDWD high-dose group (37.06 g· kg^-1· d^-1),BHDWD medial-dose group (18.53 g· kg^-1· d^-1),BHDWD low-dose group (9.26 g· kg^-1· d^-1).All the mice were administrated by intragastric with drugs or water in the same volume for 28 days.The Morris water maze was used to observe the learning and memory ability for the mice.Biochemical methods were used to determine the content of IL-6 protein in the hippocampal tissue.Results Compared with the model group,BHDWD could significantly improve the learning and memory ability of model mice (P 〈 0.05).BHDWD could obviously decrease the content of IL-6 protein in the hippocampal tissue (P 〈 0.05).Conclusion BHDWD could improve the ability of learning and memory of model mice,and its mechanism appears to involve reduction of IL-6 protein expression in the hippocampal tissue.
