The activation of Janus kinase(JAK)/signal transducer and activator transcription(STAT) pathway, which plays an important role in tumorigenesis, is aberrantly in a variety of lymphomas. Recently, the JAK/STAT inhibitors are being considered for their potentials in the therapy for lymphoma. This review summarizes the progresses on the JAK/STAT signaling pathway in the pathogenesis of lymphoma development. The genomic alterations and the mutations of JAK kinase and STATs in different lymphoma subtypes are outlined. The effects of disregulated JAK/STAT signaling and the regulatory mechanisms in lymphomagenesis are discussed. The application of JAK/STAT inhibitors as a potential therapeutic approach for lymphoma treatment is prospected.
