目的 探讨脑淋巴引流阻滞(cerebral lymphatic blockage,CLB)对蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后大鼠海马神经元凋亡的影响及相关机制研究.方法 选用健康成年Wistar大鼠,随机分为正常对照组、SAH组、SAH+CLB组.采用枕大池2次注血法建立SAH模型,于第2次注血3d后,采用HE染色及碘化丙碇(PI)染色法观察各组大鼠海马神经元形态结构变化;TUNEL荧光标记法检测原位凋亡情况;免疫组织化学激光共聚焦检测大鼠海马神经元caspase-3和Bcl-2的蛋白表达.结果 (1)HE染色和PI染色可见SAH组大鼠部分海马神经元皱缩,部分呈新月形,凋亡细胞数为(25.36 ±4.02)个;SAH+CLB组神经细胞分布稀疏,核碎裂,可见凋亡小体,周围有空泡形成,凋亡细胞数为(37.82±5.93)个,显著高于SAH组(P〈0.01).(2)SAH组和SAH+CLB组TUNEL阳性细胞的表达荧光强度分别为(1.70±0.37)和(2.54±0.53),均高于正常对照组(0.19±0.03),而SAH+CLB组又显著高于SAH组(P〈0.01).(3)SAH组和SAH+CLB组caspage-3表达的荧光强度分别为(2.45±0.49)和(2.96 ±0.44),均高于正常对照组,而SAH+CLB组又显著高于SAH组(P〈0.01).(4)SAH组和SAH+CLB组Bel-2表达的荧光强度分别为(3.40±0.61)和(2.67 ±0.44)均高于正常对照组,而SAH+CLB组显著低于SAH组(P〈0.01).结论 脑淋巴引流阻滞可加重SAH后大鼠海马神经元的凋亡,其机制可能与caspase-3高表达和Bcl-2低表达有关.
Objective To investigate the influence of cerebral lymphatic blockade (CLB) on apoptosis of hippocampal neurons after subarachnoid hemorrhage (SAH) in rats. Methods Healthy adult Wistar rats were randomly assigned to normal control group,SAH group and SAH + CLB group. SAH model was induced by double injection of autologous blood into the cistema magna. On day 3 after second injection, hippocampal cell shape structure of each group were determined by hematoxylin-eosin staining (HE) and propidium iodide (PI) staining. Terminal-deoxynucleotidy transferase mediated nick end labeling (TUNEL) fluorescent was used to determine the situ apoptosis. Immunohistochemistry was conducted to study the expression of caspase-3 and Bcl-2 in hippocampal neurons. Results (1) HE staining and PI staining showed the hippocampal neurons of SAH rats were partly shrink,and nuclei showed wavy or folded seam-like,some crescent-shaped; the hippocampal neurons in SAH + CLB group distributed sparsely,nuclear fragmentation,apoptotic bodies could be seen,surrounded by vacuole formation, Compared with the SAH group, the number of apoptotic cells in SAH + CLB group was significantly increased(the number of apoptotic cells: 0.71 ±0.05,25.36 ±4. 02,37. 82 ±5.93, P〈0.01). (2) The fluorescence intensity of positive cells by TUNEL stain in SAH group and SAH + CLB group was higher than in normal control group,while the SAH + CLB group was significantly higher than the SAH group (the fluorescence intensity: 0.19 ±0.03,1.70 ±0.37,2.54±0.53, P〈0.01). (3) The fluorescence intensity of caspase-3 in SAH group and SAH + CLB group was higher than the normal control group, while the SAH + CLB group was significantly higher than the SAH group (the fluorescence intensity: 0.14 ±0.03,2.45 ±0.49,2.96 ±0.44, P〈0.01).
