目的应用蛋白质组学研究单肺通气及急性肺损伤(ALI)/急性呼吸窘迫综合征(ARDS)的差异蛋白,筛选单肺通气潜在肺损伤相关标志物。方法检索数据库应用蛋白质组学研究ALI/ARDS生物标志物的文献(文献组);应用同位素标记绝对和相对定量(i TRAQ)技术检测20例行单肺通气肺癌患者血清差异蛋白质(实验组)。分析文献数据与实验数据的异同。结果文献组检索获得符合要求文献13篇,涉及ALI/ARDS蛋白标志物229个;实验组共鉴定出189个蛋白,至少在2个时间点发生差异表达的蛋白质31个,利用生物信息学选出11个核心蛋白。综合两组经数据分析后,共筛选出21个潜在肺损伤差异蛋白,其中有9个蛋白被重复发现2次以上,包括触珠蛋白、S100钙结合蛋白A8、抗凝血酶Ⅲ等。结论数据分析后获得的21个蛋白质可能是潜在增加单肺通气肺损伤的生物学标志物,值得进一步研究。
Objective To screen the potential markers for lung injury from one-lung ventilation by comparing and analyzing the differentially expressed proteins both from one-lung ventilation and acute lung injury/acute respiratory distress syndrome (ALI/ARDS) based on proteomics. Methods A retrieval of literatures on ALI/ARDS biomarkers of ALI/ARDS identified by proteomics (literature group ) was performed. The differential proteins in 20 serum samples which were collected from 20 lung cancer patients undergoing one-lung ventilation were detected using the technique of isobaric tags for relative and absolute quantitation ( iTRAQ ) ( experimental group ). A comparative analysis of literature data and experimental data was conducted. Results Thirteen pieces of eligible literature were obtained by information retrieval in the literature group,involving 229 ALI/ARDS biomarkers. There were 189 identified proteins in the experimental group,involving 31 proteins which presented different expressions at least at two time points, and 11 core proteins were selected using bioinformatics. Based on the data analysis of the two group,21 potential biomarkers for lung injury were selected, 11 core proteins were in the experimental group,of which 9 proteins were found more than two times including haptoglobin, S100-A8, antithrombin-Ⅲ and so on. Conclusion These 21 proteins based on data analysis might be potential biomarkers for lung injury in patients undergoing one-lung ventilation which are worth further research.
