通过研究猪毛菜(Salsola collina)对小鼠肠运动的影响,深入探讨其作用机制。建立正常小鼠肠推进模型,检测大、中、小剂量(0.025,0.05,0.1g·mL-1)猪毛菜对肠推进的影响。分别采用阿托品和肾上腺素建立小鼠肠推进减慢模型,观察不同剂量猪毛菜对肠推进的影响。通过酶联免疫吸附试验(Elisa法)分别测定猪毛菜对正常小鼠血清和肠组织中的胃动素和胃饥饿素含量的影响。实验结果表明:大、中、小剂量猪毛菜对正常小鼠肠运动具有显著促进作用(分别为p〈0.05,p〈0.01和p〈0.01),还可显著改善阿托品致小鼠肠推进减慢作用(p〈0.01),但对肾上腺素致小鼠肠推进减慢无显著影响(p〉0.05)。猪毛菜能显著提高血清中胃饥饿素含量(p〈0.01),但对肠组织中胃饥饿素含量以及血清和肠组织中的胃动素含量无显著影响(p〉0.05)。猪毛菜可通过激动M-胆碱能受体促进胃饥饿素合成,多靶点促进胃肠运动。
The effect and its mechanism of Salsola collina on intestinal motility in mice was explored.Normal mouse intestinal propulsion model was established and the influence of different dosage (0.025,0.05,0.1 g·mL-1)of Salsola collina on intestinal propulsion was detected.Prolonged intestinal propulsion of mice model induced by atropine and adrenaline was established to observe the influence of different dosage of Salsola collina on intestinal propulsion.The effects of Salsola collina on serum and intestinal motilin and ghrelin in normal mouse were detected by enzyme linked immunosorbent assay (Elisa).The results showed that three doses of Salsola collina significantly promoted intestinal motility in normal mice (p〈0.05, p〈0.01 and p〈0.01), and significantly improved the atropine induced intestinal propusion(p〈0.01), while had no effect on adrenaline induced intestinal propulsion (p〉0.05).Salsola collina can significantly improved the content of serum ghrelin (p〈0.01), but had no effect on serum and intestinal motilin and intestinal ghrelin (p〉0.05).Salsola collina is possibly promotes intestinal motility by multiple targets such as exciting M-cholinergic receptor and promote ghrelin synthesis.
