【目的】:uPA及uPAR系统在牛卵泡生长调节方面的研究会取得较大进展而有着广阔的应用前景。本实验探讨牛未成熟卵丘-卵母细胞中uPA和uPAR蛋白定位与表达。【方法】:分别应用western blot及FITC免疫荧光方法检测牛未成熟卵丘-卵母细胞中uPA和uPAR蛋白定位与表达。【结果】:Western blot及FITC免疫荧光方法检测发现uPA在牛未成熟卵丘细胞、卵泡内膜细胞和外膜细胞表达,uPA在牛卵母细胞不表达;并发现uPAR在牛未成熟卵丘细胞、卵泡内膜细胞和外膜细胞表达,uPAR在牛卵母细胞表达。【结论】:根据western blot及FITC免疫荧光方法的结果推测,uPA可通过结合牛卵丘细胞和卵母细胞上的uPAR调节卵泡的生长。
Objective: uPAs may modulate the growth of bovine follicle, study on this part may get big progression and has a wide prospect. This study examination the expression of urokinase Plasminogen activator(uPA) and urokinase Plasminogen activator recep- tor(uPAR) protein localization and expression in bovine pre -mature COCs. Method: The localization and expression of uPA and uPAR in bovine COCs were detected by western blot and FITC immunofluorescence respectively. Results : Western blot technique and the FITC immunoflnorescence technique detect uPA is expressed in the bovine pre -mature cumulus cells but not in oocyte, uPAR is expressed in the bovine pre - mature cumulus cells and oocyte. Conclusion : Depend on the resuh of Western blot technique and the FITC immunofluorescence technique, we guess uPA may modulate the growth of follicle by connect to the uPAR on bovine cumulus cells and oocyte.
