目的探究蝙蝠葛碱对雌激素替代疗法治疗卵巢切除小鼠骨质疏松症效果的增强作用。方法对3月龄雌性C57BL/6小鼠实施双侧卵巢切除手术。术后1周,对卵巢切除小鼠进行为期5周雌二醇腹腔注射或雌二醇腹腔注射联合蝙蝠葛碱灌胃治疗。治疗5周后,通过X线成像技术对小鼠股骨颈进行骨密度检测,通过HE染色对小鼠股骨颈骨小梁结构进行组织病理学检测。结果 C57BL/6小鼠接受双侧卵巢切除手术5 d后,血清雌二醇水平显著下降;术后6周,X线成像显示股骨颈部位骨密度明显降低,HE染色显示股骨颈平均骨小梁间隙(MTS)显著增加。持续5周的雌激素替代治疗可使卵巢切除小鼠股骨颈骨密度和MTS显著恢复;而蝙蝠葛碱可进一步提高雌激素治疗效果,使小鼠股骨颈骨密度和MTS恢复至卵巢功能正常同龄小鼠水平。结论蝙蝠葛碱可显著增强雌激素对卵巢切除小鼠骨质疏松症的治疗效果。

英文摘要:

Objective To investigate the enhancing effect of dauricine on the therapeutic efficacy of estrogen for the osteoporosis of ovariectomized mice. Methods Three-month old female C57BL/6 mice were subjected to the bilateral ovariectomy. The ovariectomized mice were treated with the estradiol(i.p.) or estradiol(i.p.) combined with dauricine(gavage) for 5 weeks 1 week post surgery. 5-week medication later, the bone density of the murine collum femoris was monitored by X-ray imaging and the structure of bone trabecular was evaluated by HE staining. Results The ovariectomized C57BL/6 mice showed dramatically decreased serum estradiol level 5 days post surgery and significantly reduced bone density of collum femoris, and evidently increased mean trabecular separation(MTS) 6 weeks post surgery. After the estrogen-replacement therapy lasting for 5 weeks, the bone density of collum femoris and MTS of ovariectomized mice were notably recovered. The bone density and MTS were restored to the levels of the normal mice while estradiol and dauricine were combined. Conclusion Dauricine significantly enhances the therapeutic efficacy of estrogen for the osteoporosis of ovariectomized mice.
