为了探讨产乳酸的芽孢杆菌对青贮发酵的作用。以青贮三个月的豆科牧草疏叶骆驼刺为材料,进行MRS培养基分离、纯化细菌,革兰氏染色,芽孢染色,细胞形态学观察,生理生化特性鉴定和16S rDNA序列分析。结果显示,菌株杆状,G+,有芽孢,产乳酸;16S rDNA测序结果显示该菌株与模式菌株ATCC 14580的相似性为99.3%。结果表明,该菌为地衣芽孢杆菌(Bacillus licheniformis)。
In order to explore the effect of lactic acid-producing Bacillus on silage fermentation, using Alhagi sparsifolia Shap. ensiled for three months as materials, Bacillus was isolated and purified with MRS medium. And then the isolate strains were identified with Gram staining, spore staining, cell colony morphology observation, physiological and biochemical characteristics identification and 16 S rDNA sequencing analysis were used to identify. Result showed that the bacterium were rod-shaped, Gram-positive, spore-forming, and lactic acid-producing. The 16 S rDNA result showed that the similarity between the strain and the published sequence(type strain ATCC 14580) was 99.3%. To sum up, the strain was identified as Bacillus licheniformis.
