防龋DNA疫苗pGJA—P/VAX免疫小鼠后的抗DNA抗体检测

目的:检测防龋DNA疫苗pGJA—P/VAX经鼻腔免疫和肌肉免疫后是否会产生抗DNA抗体。研究该疫苗是否会导致自身免疫性疾病。方法:提取纯化的质粒pGJA—P/VAX分别经鼻腔和肌肉免疫小鼠,于第0、2、4、8周取血清做抗DNA抗体检测实验(ELISA法)。结果:各实验组之间均无显著性差异,各组免疫前后及各组问比较(P〉0,01)。结论:说明DNA疫苗pGJA—P/VAX及其载体pVAXl对小鼠均不会引起自身免疫反应,导致自身免疫病。

英文摘要:

Objective: To compare the levels of anti -ds -DNA antibodies and anti -ss -DNA antibodies in mice immunized with pGJA - P/VAX and mice without such treatment. Methods : pGJA - P/VAX was administrated imtramuscularly or intranasally to mice, and all mice received a booster immunization 2 weeks later after the initial immunization. The concentrations of anti - ssDNA antibodies and anti - dsDNA antibodies of unimmunized mice were measured by the method of ELISA, which were used as baseline for the comparison study. 2, 4 and 8 weeks after the initial immunization, blood samples of these immunized mice were collected to check the levels of anti - ssDNA antibodies and anti - dsDNA antibodies by using the same method. Results : No statistically significant changes of anti - ssDNA antibodies and anti - ds - DNA antibodies were found between unimmunized and immunized mice. Conclusion : The blood levels of anti - ss - DNA antibodies and anti - ds - DNA antibodies did not statistically significantly increase by the repeated and high dose adminstration of anticaries DNA vaccine pGJA - P/VAX. Additionally, autoimmune diseases were found among the immunized mice at the specific experiment condition.
