利用Box-Behnken设计响应面法优化超声波辅助双水相法提取玛咖多糖的条件.采用玛咖干根为原料,考察了提取温度、提取时间、料液比3个因素对提取结果的影响.在单个变量因子实验基础上,通过采用Box-Behnken设计响应面分析法对玛咖多糖的超声波双水相提取条件进行优化.最佳提取条件为提取温度60℃、料液比1:20(质量比)、提取时间52min.与二次方程的拟合度在0.8153,得到玛咖多糖的提取率为15.831%,与实测值15.732%基本一致.Box-Behnken响应面法用于优化超声波辅助双水相法提取玛咖多糖的条件是可行的,模型预测效果较好,优化条件具备可行性.
A 17-run Box-Behnken design was used to optimize the extraction conditions of polysaccha- rides from maca via ultrasonic-assisted aqueous two-phase method.Three factors, namely, extraction tem- perature,extraction time, and solid-liquid ratio were investigated. The experimental data were fitted to a second-order polynomial equation through multiple regression analysis and examined with the appropriate statistical methods. The adjusted coefficient of determination (RAdj2)for the model was 0.815 3, and the probability value (P〈0.000 1)demonstrated a high significance for the regression model.The optimum ex- traction conditions were as follows: extraction temperature of 60 ℃, extraction time of 52 min, and solid- liquid ratio of 1 : 20. Under these conditions,the mean extraction yield of maea polysaecharides was 15. 732% ,which was in good agreement with the predicted value of 15. 831%. Box-Behnken response surface method is used to optimize extraction of polysaccharides from maca via ultrasonic-assisted aqueous two- phase method, the optimized conditions are feasible, the model predition is good.
