目的:探讨烫伤后创面脓毒症大鼠肝型肉碱棕榈酰转移酶1(liver carnitine palmitoyltransferase 1,L-CPT1)mRNA的表达规律及意义.方法:60只背部30%Ⅲ度烫伤大鼠随机分为创面脓毒症组和烫伤对照组,创面脓毒症组大鼠烫伤后10 min创面涂布绿脓杆菌,建立创面脓毒症模型,通过实时荧光定量PCR法测定伤前、伤后24h、48 h、96 h及7 d的L-CPT1 mRNA表达变化.结果:大鼠烫伤后肝组织L-CPT1 mRNA的表达在伤后早期轻度增高,此后表达明显下调,创面脓毒症组下降趋势更明显,致伤后96 h表达量降低至最低点,并显著低于对照组(P<0.05).结论:烫伤后创面脓毒症的发生引起肝脏L-CPT 1 mRNA表达下调,导致脂肪酸活化产物脂酰辅酶A向线粒体内转运减少,加重了烫伤后的脂代谢紊乱.
Objective :To investigate the expression of liver carnitine palmitoyltransferase 1 ( L-CPT 1 ) mRNA in liver of scalded rats with wound sepsis. Methods: 60 male Wistar rats were scalded resulting in 30% TBSA full-thickness bum and then randomly divided into wound sepsis group and control group. Pseudomonas aeruginosa (10^9/ml) was inoculated to the rats' wounds of sepsis group. The quantity of bacteria in the subeschar tissue was mensurated to confirmed that the rat model of burn wound sepsis was established successfully. Fresh liver tissues were obtained. Total RNA of liver was isolated with Trizol and the levels of L-CPT 1 were measured by reverse transcription polymerase chain reaction (RT-PCR). Results:Compared with control group, L-CPT 1 mRNA levels of sepsis group were significantly decreased in liver during 24 - 96h post scalding (P 〈 0.05 ). Conclusions:Burn wound sepsis can decrease the expression of L-CPT 1 mRNA in liver, which might decreased the entry of long-chain fatty acids into the mitochondrial matrix, and involved in the development of lipid metabolism disturbance.
