目的:探讨益气化瘀方对椎间盘软骨细胞凋亡状态下Bax、Bcl-2及Caspase-8表达的调控作用。方法:清洁级雄性1月龄SD大鼠20只,酶消化法获取椎间盘软骨细胞,用抗Fas抗体诱导其发生凋亡,通过益气化瘀方及其拆方益气方、化瘀方药理血清干预,免疫组化法(SP)检测分析大鼠椎间盘软骨细胞不同状态下Bax、Bcl-2及Caspase-8的表达。结果:相对正常组,凋亡模型组的Bax和Caspase-8表达明显,而Bcl-2轻度表达,有显著差异(P〈0.01)。益气化瘀方、益气方和化瘀方均能降低椎间盘软骨细胞的Bax、Caspase-8的表达,而上调Bcl-2的表达,3种中药组与诱导凋亡组相比有显著差异(P〈0.01)。结论:中药益气化瘀方、益气方和化瘀方都可以调控凋亡的大鼠椎间盘软骨细胞内Bax、Bcl-2、Caspase-8的表达,其治疗颈椎病的机制与其对凋亡相关因子的调控有关。
Objective: To study the effects of Yiqi-Huayu recipe (益气化瘀方) and its decomposed formulas-medicated on expressions of Bcl-2, Bax and Caspase-8 of apoptotic discus intervertebrale chondrocytes in rats. Methods: Immunohistochemical and integral optical density analytic methods were used to observe the effects of Yiqi-Huayu recipe, Yiqi recipe, Huayu recipe-medicated sera on the expressions of Bcl-2, Bax and Caspase-8 of apoptotic discus intervertebrale chondrocytes in rats induced by anti-Fas antibody. Results: As compared with apoptosis group, Bcl-2 expression was higher, Bax and Caspase-8 expressions were lower in Yiqi-Huayu-treated, Yiqi-treated, Huayu-treated ( P 〈 0.01 ). Conclusion: Yiqi-Huayu recipe and its decomposed formulas can delay degeneration of the cervical intervertebral disc, which may be due to its action in regulating the expressions of Bcl-2, Bax and Caspase-8.
