目的 探讨miR-145对口腔癌细胞增殖、迁移、侵袭生物学特性的影响。方法 运用脂质体介导的转染方法将miR-145转染入口腔癌细胞TCA8113,通过细胞增殖实验检测转染后口腔癌细胞TCA8113增殖生物学特性的改变,通过细胞迁移实验、细胞侵袭实验,检测口腔癌细胞TCA8113细胞迁移、侵袭生物学特性的变化。结果 miR-145转染组细胞增殖活性明显低于对照组(P〈0.05),细胞迁移、侵袭活性明显低于对照组(P〈0.05)。结论 miR-145的表达参与口腔细胞癌的发生及发展过程,miR-145的高表达可对口腔癌细胞TCA8113发挥负性调控作用。
Objective To investigate the effects of miR-145 on the biological characteristics including proliferation, migration and invasion of oral carcinoma cell TCA8113. Methods The lipofectamine transfection method was used to improve the miR-145 expression in oral carcinoma TCA8113. The biological characteristics changes of oral carcinoma cell TCA8113 in vitro were assayed by proliferation test, migration test, and invasion test. Results After miR-145 transfection, the proliferation, migration and invasion characteristics of oral carcinoma TCA8113 were markedly decreased compared with those in control group (P〈0.05). Conclusion The expression of miR-145 is associated with the genesis and development of oral carcinoma. Overexpression of miR-145 can negatively regulate oral carcinoma TCA8113.
