目的:探讨黄连素(berberine)对巨噬细胞清道夫受体CD36、LOX-1、CD68等表达的影响。方法:以佛波醇酯(PMA)诱导分化的单核源巨噬细胞为细胞模型,黄连素(50μmol/L)预处理1 h后,加入氧化低密度脂蛋白(oxLDL)刺激,采用流式细胞仪检测巨噬细胞表面清道夫受体的表达。结果:单核细胞分化成巨噬细胞后,CD36表达明显升高,而黄连素预处理显著抑制CD36的表达。oxLDL刺激巨噬细胞24 h后,巨噬细胞表面清道夫受体CD36、LOX-1和CD68表达明显升高(P〈0.01),黄连素预处理后巨噬细胞表面CD36、LOX-1的表达受到抑制,但CD68的表达未受影响(P〉0.05)。结论:黄连素抑制巨噬细胞表面清道夫受体CD36、LOX-1的表达,一定程度上可减少巨噬细胞通过清道夫受体途径对脂质oxLDL的吞噬。
Objective: To investigate the effect of berberine on the expression of scavenger receptors(CD36,LOX-1,CD68) in macrophages.Methods: PMA-induced macrophages stimulated by oxLDL was used as model in this study to investigate the expression of scavenger receptors,including CD36,CD68 and LOX-1,which was tested using Flow cytometry.Results: Berberine greatly inhibited CD36 expression during monocytes differentiation to macrophages and similar result was obtained in oxLDL-stimulated macrophages.Moreover,in oxLDL-stimu-lated macrophages,LOX-1 expression,but not CD68,was also decreased after berberine treat-ment than those in oxLDL only group.Conclusion: Berberine suppresses the expression of CD36 and LOX-1,but not CD68 in macrophages.
