目的:探讨脂联素基因rs864265多态性与动脉粥样硬化的关系,评估代谢危险因素与rs864265多态性交互作用对动脉粥样硬化的影响。方法以来自江苏徐州地区的1609名健康体检人群为研究对象,运用实时荧光定量PCR技术对rs864265多态性进行基因分型,并检测参与者的动脉脉搏波传导速度及代谢因素相关指标,进行统计分析。结果校正年龄及性别因素后,T 等位基因与颈-股动脉脉搏波传导速度(cfPWV)呈正相关性(趋势P=0.001)。进一步校正体质指数、血压、空腹血糖和脂质后,这个趋势没有明显改变(趋势P=0.012)。rs864265多态性与年龄、体质指数、体脂率、腰围及收缩压交互作用对动脉粥样硬化的影响有统计学意义(交互作用 P 值分别为0.031、0.035、0.001、0.003、0.013)。结论 rs864265多态性与动脉粥样硬化相关联,并独立于其他代谢危险因素。年龄、体质指数、体脂率、腰围及收缩压可影响这种相关性。
Objective To investigate the associations of rs864265 polymorphism in the adiponectin gene (ADIPOQ) with atherosclerosis, and particularly assess the interactions between the genetic polymorphism and metabolic risk factors. Methods The study samples were from a community-based health examination survey in Xuzhou, Jiangsu province of China. A total of 1 609 subjects were included in the present study. rs864265 polymorphism was genotyped by real-time PCR, pulse wave velocity (PWV) and metabolic biomarkers features were evaluated, and the statistical analysis was made. Results After adjustment for age and sex, carriers of rs864265 T allele had significantly greater increased carotid femoral PWV (cfPWV) (P for trend=0.001). Further adjustment for body mass index, blood pressure, fasting blood glucose and lipids did not significantly change the result (P for trend=0.012). We found significant interactions between this genetic polymorphism and age, body mass index, body fat rate, waist circumference and systolic blood pressure in relation to cfPWV (P for interaction=0.031, 0.035, 0.001, 0.003, 0.013, respectively). Conclusions rs864265 polymorphism was significantly associated with atherosclerosis, independent of other metabolic risk factors. The association may be amplified by age, body mass index, body fat rate, waist circumference and systolic blood pressure.
