目的研究hS100A6(human S100A6)对人卵巢癌细胞系(低转移系HO8910和高转移系HO8910 PM)增殖、凋亡和Wnt/β-catenin信号途径的影响。方法通过MTT以及台盼蓝染色检测细胞增殖的变化;Hoechst检测细胞凋亡的变化;提取细胞总蛋白及核蛋白Western blot检测hS100A6对β-catenin的影响。结果 MTT及台盼蓝染色结果显示hS100A6抑制HO8910细胞和HO8910 PM细胞的增殖(P〈0.05),Hoechst染色结果显示hS100A6同时促进这两系细胞的凋亡(P〈0.05);Western blot显示hS100A6增加β-catenin的水平并促进其发生核转位。结论 hS100A6能抑制人卵巢癌细胞系HO8910和HO8910 PM增殖,促进其凋亡,同时激活Wnt/β-catenin信号途径。
Objective To investigate the effect of human S100A6(hS100A6) on the proliferation,apoptosis and Wnt/β-catenin signaling pathway of ovarian cancer cell line HO8910 and HO8910 PM.Methods MTT assay and trypan blue exclusion assay,Hoechst staining and Western blotting were used to detect cell proliferation,apoptosis and expression and distribution of β-catenin,respectively.Results ① hS100A6 inhibited proliferation of HO8910 and HO8910 PM cells in a dose-and time-dependent manner(P0.05);② hS100A6 induced apoptosis in HO8910 and HO8910 PM cells(P0.05);③ Level of β-catenin was increased by hS100A6 in HO8910 and HO8910 PM cells,and β-catenin translocated from cytoplasm to nucleus.Conclusion hS100A6 can inhibit cell proliferation,induce cell apoptosis and up-regulate Wnt/β-catenin activity in ovarian cancer HO8910 and HO8910 PM cells.
