Phospholipase A2 （PLA2）, which exists widespreadly in the mammalian, is one of the most crucial enzymes that play important roles in cell metabolism, signal transduction and so forth. With the further research of phospholipase A2, more than 30 enzymes that possess phospholipase A2 or related activity have been identified in mammals and it helps to lead a new advance in revealing the male reproduction relevant phospholi- pase A2 isozymes. This review focuses on the current understanding of the classification and characterization of phospholipase A2 family members, the localizations ofphospholipase A2s in male reproductive system, the roles of phospholipase A2 and its signal transduction pathways in the acrosome reaction, phospholipase A2 subtypes relevant to the male reproduction, etc. This article is a study on the roles and the mechanisms ofphospholipase A2s in male reproductive system which may provide a theoretical basis and thinking for a strategy in the male infertility diagno- sis and treatment.
