目的探讨不同比例肝叶切除术后门静脉压力(PVP)变化与力学信号传导通路关键蛋白黏着斑激酶(FAK)表达及肝再生的关系。方法将64只Sprague-Dawley雌性大鼠随机分为假手术组、70%肝叶切除术组、50%肝叶切除术组和30%肝叶切除术组。测定术后24h、48h、72h和168h门静脉压力值(每组每个时点各4只)。免疫组化方法检测肝组织FAK和细胞核增殖抗原(PCNA)表达,并用hnage-pro plus6.0医学图像分析软件进行半定量分析。结果肝叶切除比例越大,相应时点门静脉血流压力值越大,肝组织FAK和PCNA表达越强。术后24h、48h、72h和168h各时点PVP、FAK以及PCNA水平三者呈正相关。结论肝切除术后门静脉压力升高可通过激活力学信号传导通路关键蛋白FAK促进肝细胞增殖和肝组织再生。
Objective To explore discuss the relationship of the portal venous pressure (PVP) with the expression of focal adhesion kinase ( FAK), which is the key protein of mechanics related signaling pathways, and liver regeneration in partial liver resection models. Methods 64 female Sprague-Dawley rats were randomly divided into 4 groups : sham-operation group, 70% liver resection group, 50% liver resection group and 30% liver resection group. PVP was observed in 24 h, 48 h, 72 h and 168 h after liver resection ( n = 4 each group at each time point). FAK and PCNA levels .were detected in remaining liver by immunohistochemical staining method, Image-pro plus 6.0 Digimizer was used to conduct the semi-quantitative analysis. Results As more liver was reseeted, the PVP value and FAK and PCNA expression were increased accordingly at each time point. In 24 h, 48 h and 72 h after surgery, PVP was positively correlated with FAK and PCNA level. Conclusion After liver resection, the increased PVP may promote liver cells proliferation and liver regeneration through activating FAK signaling.
