目的研究T细胞因子3（T cell factor 3,TCF-3）在非小细胞肺癌中的表达及分布情况。方法采用50对人非小细胞肺癌组织标本及其正常癌旁组织对照,进行免疫组化检测TCF-3蛋白在肺癌组织及正常肺组织中的表达及分布情况;采用5种人非小细胞肺癌细胞系及2种正常人支气管上皮细胞系进行实时荧光定量PCR,Western blot检测肺癌细胞系中TCF-3在mRNA和蛋白水平的表达情况。结果 （1）TCF-3在肺癌组织中表达,表达率为48%（24/50）,在正常肺组织中未见表达。TCF-3在肺癌细胞胞核与胞质同表达,以细胞核表达为主。（2）TCF-3在A549、H520、HBE、BEP-2D 4种细胞系中mRNA及蛋白水平均有表达。结论 TCF-3在肺癌组织及细胞系中均表达,TCF-3蛋白在细胞核与细胞质同表达,但以细胞核表达为主。

Objective To determine the expression profiles and distribution of T cell factor-3（ TCF-3）in non-small cell lung cancer（ NSCLC）. Methods TCF-3 expression was examined in 50 NSCLC tissue specimens and their corresponding normal lung specimens by immunohistochemical assay（ S-P method）. The mRNA levels of TCF-3 were detected in 5 cell lines of NSCLC（ A549,H460,H226,H520 and SK-MES-1cell lines） and 2 human normal bronchial epithelial cell lines（ HBE and BEP-2D cell lines） by real-time fluorescence quantitative PCR. The protein level of TCF-3 gene in the cell lines was detected by Western blotting. Results Among the 50 lung cancer specimens,24 samples showed high expression of TCF-3（ 48%）,and 22 samples had the expression both in the cytoplasm and nuclei. But no TCF-3 expression was detected in 50 corresponding normal lung specimens. The expression of TCF-3 was found at mRNA and protein levels in the cell lines A549,H520,HBE,and BEP-2D. Conclusion TCF-3 is expressed in the NSCLC tissue as well as cell lines. TCF-3 protein is located in the nuclei and cytoplasm,but mainly in the former.
