试验以荒漠区药用植物黑果枸杞无菌苗的子叶、茎段、胚轴为外植体材料,研究不同激素配比的培养基对不同外植体愈伤组织培养、丛生芽增殖及生根培养的影响,并筛选适宜各个阶段培养的最佳培养基配方。结果表明:子叶是诱导黑果枸杞愈伤组织的最好外植体,在6-BA为0.3mg/L,2,4-D为1.0mg/L时,其愈伤组织诱导率达到最大值95.1%;较高浓度的细胞分裂6-BA对丛生芽增殖具有明显的促进作用,在6-BA为3.0mg/L、KT 0.3mg/L、NAA 0.5mg/L时丛生芽增殖率达到最大值90.2%,且丛生芽数量多、茎干健壮、生长旺盛;在以1/2MS为基本培养基,减少无机盐的浓度时对黑果枸杞丛生芽生根培养具有促进作用,减少Mg SO4一半,并加入NAA0.5mg/L时,丛生芽生根率达到最大值98.2%,且生根时间最早,主根明显、粗壮且根毛数量多。

英文摘要:

The experiments using the cotyledons, hypocotyl explants and stem section of Lycium ruthencium as material to research influence of different ratio of hormone on different explant callus culture, the multiple shoot clumps proliferation and rooting culture, and select the best culture medium formula suitable for all stages. The results show that the cotyledon is the best explant to callus induction. While 6 - BA was 0.3mg/L and 2, 4 - D was 1.0mg/L, the callus induction rate reached a maximum of 95.1%. The high levels of cell division 6 - BA had obviously promoting effect on multiple shoot clumps proliferation. When 6 - BA was 3.0mg/L, KT 0.3mg/ L, NAA 0.5mg/L multiple shoot clumps proliferation rate reached a maximum of 90.2% , and the multiple shoot clumps number was large, stem was robust with vigorous growth. Using 1/2 MS as the basic culture medium, and reducing the concentration of inorganic salt had a promoting effects on the Lycium ruthencium multiple shoot clumps growth, reducing half of MgSO4 and joining NAA 0.5mg/L, the multiple shoot clumps maximum rooting rate reached 98.2%, and began to take root the earliest time, and taproot was strong and root hairs were thick.
