以总黄酮回收率为指标,研究了大孔树脂分离纯化核桃叶总黄酮的工艺。结果表明:LS303型树脂对核桃叶总黄酮有较好的吸附分离性能,是分离纯化核桃叶总黄酮的适宜大孔树脂;分离纯化核桃叶总黄酮的最佳工艺条件为:黄酮量与干树脂量比为1:8,浓度为4.67mg·mL^-1,pH6,4BV的水洗后用4BV的70%乙醇洗脱。经LS303处理后的核桃叶总黄酮回收率在80%以上,纯度70%以上。该工艺简便,能有效分离纯化核桃叶黄酮化合物。
A method for separation and purification of flavonoids from walnut leaves with macroporous resin was studied, with the recovery rate of flavonoids as index. The results were as flollows: among six types of macroporous resin choosed, resin LS303 was the best one. The optimum process conditions were the loaded amount of flavonoids vs dry resin: 1 : 8, the extract solution at a concentration about 4.67 mg · mL^-1 , pH 6. After eluted with 4BV of distilled water and 4BV of 70% ethanol, the yield of flavonoids was above 80% and product purity was above 70%. The process is simple, the flavonoids of walnut leaves can be separated and purified effectively.
