目的:比较磷酸化抗原(HDMAPP)、结核杆菌耐热抗原(Mtb-HAg)和结核杆菌低分子耐热抗原Mtb-HAg(10 k)刺激外周血T细胞产生γ-干扰素(IFN-γ)的反应能力方面的特点及差异.方法:采集正常人外周血,分别用HDMAPP、Mtb-HAg和Mtb-HAg(10 k)刺激培养14 h,再加入莫能霉素继续培养6h,收集细胞,加入荧光标记单抗进行表面分子和胞内染色,然后在流式细胞仪上检测各实验组中CD3+ αβ细胞和CD3+ γδ细胞胞内IFN-γ的表达.结果:正常人各抗原实验组CD3+ αβ细胞和CD3+γδ细胞胞内产生IFN-γ+细胞比率较阴性组对照组升高(P<0.01).各抗原刺激组CD3+γδ细胞产生IFN-γ+细胞比率明显高于CD3+αβ细胞(P <0.05 ~P<0.01).3种抗原刺激组间CD3+αβ细胞和CD3+γδ细胞无明显不同(P>0.05).结论:HDMAPP、Mtb-HAg和Mtb-HAg(10 k)均可以刺激外周血中T细胞中αβ和γδ细胞群产生IFN-γ,均可特异性诱导刺激CD3+γδ细胞产生IFN-γ,HDMAPP、Mtb-HAg(10 k)对CD3+γδ细胞产生IFN-γ的表达量高于Mtb-HAg.
Objective: To compare the characteristics and differences of peripheral blood T cells stimulated by HDMAPP, Mycobacterium tuberculosis heat-resistant antigen (Mtb-HAg) and low molecular peptide antigen of Mycobacterium tuberculosis (Mth) (10 k) to produce interferon(IFN-γ). Methods:The peripheral blood cells from healthy adults were cultured with HDMAPP and Mtb- HAg and Mtb-HAg( 10 k) for 14 hours, then added Monensin to cultivate for another 6 hours. Cells were collected, and the surface molecule and intracellular cytokines were stained with different fluorochrome-conjugated mAbs. The expressions of IFN-γ, in CD3 + αβ and CD3 + γδ cells of different experiment group were detected by flow cytometry. Results:The ratios of CD3 αβ and CD3 + γδ cells to produce IFN-γ, in all antigen experiment groups were higher than that in control group ( P 〈 0.01 ). The ratios of CD3 γδ cells to produce IFN-γ in all antigen experiment groups were significantly higher than that of CD3 + αβ cells ( P 〈 0.05 to P 〈 0.01 ). The differences of CD3 + αβ cells and CD3 +γδ cells in three kinds of antigenic stimulation groups were not statistical significance ( P 〉 0.05 ). Conclusions:CD3 + αβ cells and CD3 + γδ cells can produce IFN-γ by stimulating the peripheral blood T cells with HDMAPP, Mtb-HAg and Mtb-HAg( 10 k). CD3 + γδ cells stimulated by three kinds of antigens can produce IFN-7. The IFN-γ level in CD3 +γ cells stimulated by HDMAPP and Mtb-HAg( 10 k) is higher than that of Mtb-HAg stimulating.
