目的研究环磷酰胺对小鼠支持细胞的影响。方法睾丸组织切片的苏木素-伊红(HE)染色观察环磷酰胺对睾丸支持细胞的影响,通过免疫组织化学和细胞化学分析检测支持细胞中CK-18等的表达,通过Western-blot法检测ERK1/2的表达。结果环磷酰胺处理导致小鼠支持细胞凋亡和数量减少,残存的支持细胞去分化,去分化的支持细胞中的ERK1/2-MAPK信号通路被激活。结论环磷酰胺处理活化了ERK1/2-MAPK信号通路并诱导支持细胞去分化。
Objective To study the effects of cyclophosphamide on Sertoli cells. Methods The effects of cyclophosphamide on Sertoli cells were observed by HE staining of mouse testis slices, the CK-18 expressions were analyzed by immunocytochemistry and immunohistochemistry, and the expressions of ERK1/2 in testis after cyclophosphamide treatment were detected by western-bloting. Results Cyclophosphamide treatment induced the apoptosis of Sertoli cells and cut down the number of Sertoli cells in testis, the remnants of Sertoli cells in mouse testis dedifferentiated, and the ERK1/2-MAPK signaling pathway was activated. Conclusion The cyclophosphamide treatment activated the ERK1/2-MAPK pathway and induced the dedifferentiation of Sertoli cells.
