目的探讨低氧(1%氧浓度)对脂肪来源干细胞增殖活性及表面标志的影响。方法用酶消化法培养脂肪来源干细胞;低氧预处理脂肪来源干细胞于1%氧浓度下48h后检测脂肪来源干细胞的增殖情况及MTT法检测处理后(24、48、72h)的细胞活性,流式细胞仪检测细胞表面标志物。结果低氧预处理期间不仅细胞增殖能力增强,而且低氧预处理后的脂肪来源干细胞具有较常氧组培养更强的活性,表面标志较常氧组无明显差异。结论缺氧预处理能提高脂肪来源干细胞的增殖能力及活性,维持脂肪来源干细胞的间充质干细胞特性,为细胞疗法的临床应用提供了新的思路。
Objective To study the effects of hypoxic pretreatment ( 1% 02 ) on the activities and cell surface marker of human adipose-derived stem cells. Methods The method of enzymatic digestion was em- ployed to isolate and culture adipose-derived stem cells. After hypoxia pretreated ( 1% 02 ) for 48 h, cell num- ber was counted, MTY method was applied to evaluate the cytoactivities at 24, 48, 72 h and the flow cytometry was used to detect cell surface markers. Results During hypoxic pretreatment, not only cell proliferation en- hanced, and the cytoactivities in hypoxia pretreatment group was stronger than that in the normoxia group, but there was no significant difference in surface markers between the two groups. Conclusion Hypoxic pretreat- ment can improve the proliferation and activity of adipose-derived stem cells with keeping mesenchymal stem cell characteristics of adipose-derived stem cells. This may provide a new thread for the chnical use of cell therapy.
