ERR1突变体K244A对其转录活化功能的影响

目的分析ERR1突变体K244A对ERR1转录活化功能的影响。方法用重叠延伸PCR构建ERR1的缺失突变体ERR1/K244A到表达质粒pSG5上.通过荧光素酶报告基因表达系统检测ERR1 K244A对ERR1转录活化功能的影响结果测序证实成功构建了ERR1的突变体pSG5/ERR1/K244A。报告基因表达系统检测显示ERR1 K244A激活的荧光素酶报告基因活性明显低于野生型的ERR1。结论ERR1突变体ERR1 K244A转录活化功能明显低于野生型的ERR1.为进一步研究ERR1的结柏与功能打下了基础.

英文摘要:

Objective To analyse the effect of mutant K244A on the activation of gene transcription of estrogen receptor related receptor α1(ERR1).Methods Mutant of ERR1 was obtained by overlap extension PCR.and then inserted into MCS of plasmid pSGS. After identification of the positive clone by digestion of restriction endonuclcase and sequcncing, the activation of luciferasc was detected by Luciferase Assay System. Results pSG5 ERR1 K244A .mutants of ERR1 ,was successfully constructed, h was found that the luciferase activity of reporter gene generated by ERR1 mutation ERR1 K244A was obviously lower vs ERR1 wild type. Conclusion The function of transcription activated by ERR1 mutation K244A was lower than ERR1 wild type . which may be the basc for further studying the constitution and ruction of ERR1.
