目的通过比较银屑病患者与对照组骨髓基质细胞分泌表皮生长因子(EGF)、血小板源性生长因子-A(PDGF-A)和血小板源性生长因子-B(PDGF-B)水平的差异,揭示银屑病患者骨髓造血微环境的异常。方法采用密度梯度离心法分离患者与对照组骨髓单一核细胞,通过贴壁法培养骨髓基质细胞,收集传3代后又培养72h的骨髓基质细胞及培养上清,用流式细胞仪鉴定细胞表面标志并用ELISA法检测上清液中EGF、PDGF-A和PDGF-B的水平。结果90%以上细胞表面抗原高表达CD29,而CD34、CD45及HLA-DR表达阴性,即骨髓基质细胞纯度在90%以上;患者组骨髓基质细胞分泌EGF和PDGF—B均显著低于对照组(P〈0.05),而PDGF—A的分泌水平两组间无统计学差异(P〉0.05)。结论银屑病患者骨髓基质细胞分泌的某些细胞因子存在异常,表明患者骨髓造血微环境可能存在异常。
Objective To reveal the abnormity of bone marrow microenvironment of psoriasis patients by comparing the level of EGF,PDGF-A and PDGF-B secreted by bone marrow stromal cells in patients and control.Methods Bone marrow mononuclear cells of patients and control by density gradient centrifuge were separated.Bone marrow stromal cells were cultured with the method of adherent.When the cells were cultured for three passages and 72 h,bone marrow stromal cells and supernatant liquid were collected.The phenotypes of cells were identified by flow cytometry and the level of EGF,PDGF-A and PDGF-B were detected by ELISA kits.Results The purity of bone marrow stromal cells was over 90%.The level of EGF and PDGF-B in patients was significantly lower than that in control(P〈0.05).And the level of PDGF-A was no significant difference between the two groups(P〉0.05).Conclusion Some of the cytokines were abnormal which secreted by psoriatic bone marrow stromal cells.Which showed that psoriatic bone marrow microenvironment may be abnormal.
