活血化瘀中药对糖尿病创面肉芽组织细胞周期变化的影响

目的:探讨活血化瘀中药对糖尿病创面肉芽组织中细胞周期的影响,探求辨证用药规律。方法:36只SD大鼠创面造模,随机分成糖尿病创面中药治疗组、糖尿病创面对照组和正常创面对照组。分别于创面修复3d、11d取材,流式细胞仪检测创面肉芽组织中细胞周期变化。结果:创伤后早期3d,糖尿病创面中药治疗组S期明显高于糖尿病创面对照组(P〈0.05),但低于正常创面对照组(P〈0.05)。创伤后11d比较, 糖尿病创面中药治疗组新生肉芽组织中S期高于糖尿病创面对照组(P〈0.05),与正常创面组无差异(P〉0.05)。而G2-M期比较,3组无统计学差异(P〉0.05)。结论:活血化瘀中药在创伤早期促使细胞增殖进入S、G2-M期,"祛瘀利于生肌",有利于创面修复。创面愈合晚期,活血化瘀中药虽能促使细胞DNA合成,但细胞增殖能力已减弱,对防止创面瘢痕过度生成有利。

英文摘要:

To study the effects of blood-promoting and stasis-removing herbs on the generation cycle of granulation tissue of diabetic rat wound healing Methods Thirty-six male Sprague-Dawley(SD) diabetic rats with fullthickness skin lesion were randomized into 3 groups: Huayu(化瘀) Recipe-treated group, diabetic untreated group and nomml NS untreated control group. The generation cycle in granulation tissue of the rats were tested with flow cytometry. Results On the third day of wound healing, cell in S stage of the rats in Huayu recipe-treated group were higher than those in the diabetic untreated group(P 〈0.05),but lower than those in the normal group (P 〈0.05).On the eleventh day of wound healing, cells in S stage of the rats in Huayu Recipe-treated group were higher than those in the diabetic untreated group ( P 〈 0.05 ) . But there was no statistical difference between these three groups ( P 〉 0.05 ) . Conclusion At the earlier period of wound healing, Huayu Recipe can promote cells of granulation tissue to proliferate into S and G2-M stages. At the advanced stage, though Huayu Recipe can promote to synthesize DNA, the ability of cell proliferation was weakened, favorable to avoid forming cicatricles.
