目的探讨密骨打老儿丸(淫羊藿、骨碎补、菟丝子、补骨脂等)防治去卵巢大鼠骨丢失和骨质量降低的作用。方法60只雌性大鼠,随机分为5组:假手术组、模型组、雌二醇组和密骨打老儿丸低、高剂量组。假手术组仅行假手术,其余4组行卵巢切除术。术后1周分别灌胃给予17β-雌二醇和密骨打老儿丸低、高剂量,连续给药3个月。测定血钙、血磷和血清碱性磷酸酶(ALP)以及尿钙(u—Ca)、尿磷、尿脱氧吡啶酚(D—Pyr)和尿肌酐(Cr)。之后处死动物,取出股骨、腰椎和胫骨,分别测定股骨密度,骨钙、骨磷,腰椎生物力学性能,胫骨组织形态计量学参数。结果与假手术组相比,模型组血清ALP、u—Ca、D—Pyr/Cr显著增加,股骨密度、腰椎生物力学参数(弹性载荷、最大载荷、断裂载荷、刚度)、胫骨上段静态参数(包括骨组织总面积、骨小梁面积、骨小梁周长)、动态参数(标记周长百分数、骨形成率)均降低。与模型组比较,密骨打老儿丸低、高剂量和雌二醇组均可使血清ALP、u—Ca、D—Pyr/Cr排出量降低,股骨密度、腰椎生物力学参数、胫骨上段静态参数、动态参数均增加。结论密骨打老儿丸能抑制由于去卵巢雌激素缺乏引发的骨转换增强,提高骨密度,增强腰椎抗压生物力学性能。
AIM To explore the influence of Migu Dalao'er Pills (Epimedii Herba, Drynariae Rhizoma, Cuscutae Semen, Psoraleae Fructus, etc. ) on mineral density, biomechanical properties and histomorphometry in ovari- ectomized osteoporosis model rats. METHODS Sixty SD female rats were randomly divided into 5 groups: sham group, model group, 17 β-estrogen treated and Migu Dalao'er Pills treated groups (2.5 mL/kg, 5 mL/kg). The sham group was only sham operated and the other four groups were ovariectomized respectively. One week after ovariectomy, the rats were given Migu Dalao'er Pills (2.5 mL/kg, 5 mL/kg) and 17β-estrogen (2 mg/kg) by in- tragastric administration for three months sustainedly. Serum contents of calcium, phosphorus, alkaline phosphata (ALP) and levels of urinary deoxypyridinoline (D-Pyr) , urine contents of calcium (u-Ca), phosphorus and creatinine (Cr) were detected. Rats were sacrificed at the end of the experiment. The thighbone was taken out to determine bone mineral density (BMD). The second lumbar vertebra was taken out to determine contents of calcium and phosphorus. The forth lumbar vertebrae was taken out to measure biomechanics properties. The proximal tibia was taken out to determine bone histomorphometric parameters. RESULTS Compared with the sham group, serum ALP activity as well as u-Ca and urinary D-Pyr/Cr significantly increased in ovariectomized rats. BMD of femur, biomechanical properties (elastic load, maximum load, break load, stiffness), static parameters (including total tissue area, trabecular area, trabecular perimeter) and dynamic parameters (% L. Pm, BFR/BV and BFR/ TV) significantly decreased in the model group. Compared with the model group, high and low doses of Migu Dalao'er Pills groups and 17β-estrogen group could make serum ALP activity inhibit, and u-Ca, urinary D-Pyr/Cr reduce.
