目的观察电针丰隆穴对高脂血症大鼠ABCA1mRNA表达的影响。方法36只健康雄性SD大鼠随机分为空白组、模型组、电针组,每组12只。用高脂饮食饲料饲喂法建立高脂血症大鼠模型。造模28d后.电针组开始电针丰隆穴治疗。治疗结束后,检测各组大鼠血脂,并用原位杂交法检测各组大鼠肝脏及主动脉弓组织中ABCA1mRNA表达水平。结果与空白组相比,模型组大鼠TC、LDL—C水平显著上升,肝脏及主动脉弓ATP结合盒转运子A1(ABCA1)mRNA表达显著降低;与模型组比较,电针组大鼠TC、LDL—C水平显著降低,肝脏及主动脉弓ABCA1mRNA表达升高。结论电针丰隆穴能够增强高脂血症大鼠肝脏及主动脉弓ABCA1mRNA的表达,从而促进胆固醇的逆向转运,这可能是电针丰隆穴降脂及防治动脉粥样硬化的机制之一.
Objective: To investigate the effect and mechanism of electroa-cupuncture (EA) at Fenglong (ST 40) on ABCAlmRNA in hyperlipidemia (HLP) rats. Methods: 36 healthy SD rats were randomly divided into normal control group,model group and EA group. Modeling after 28 days,the EA groups were given EA Treatment. At the end of the treatment, the lipids and expression of ATP binding cassette transporter A 1 (ABCA 1 ) mRNA in liver and aortic arch tissue were assayed by in situ hybridization (ISH). Results: The content of TC and LDL-C of the model group were apparently increased and the expression of ABCA1 mRNA were significantly decreased compared with the nomad control group ; compared with the model group,levels of TC and LDL-C were significantly decreased and the expression of ABCA1 mRNA were significantly increased in the the EA group. Conclusion: Electro-acupuncture at the acupoint Fenglong can promote reverse cholesterol transport through improving the expression of ABCA1 mRNA ,which may be one of the mechanisms of EA at fenglong lipid-lowering and preventing AS.
