骨科内植物纳米涂层抗菌性能研究进展

目的 探讨姜黄素对大鼠胶原诱导性关节炎(CIA)的治疗效果及作用机制.方法 雄性无特定病原体(SPF) Wistar大鼠45只随机分配为3组(A组为空白对照组,B组为CIA模型组,C组为CIA模型姜黄素治疗组),每组15只.B组和C组大鼠经胶原蛋白诱导构建CIA模型.实验第21天起给予C组大鼠姜黄素灌胃治疗(2天1次),每次给药后观察记录大鼠关节炎评分.治疗前及治疗结束后分别摄X线片并予影像学评分.治疗2周后处死动物行常规苏木精-伊红(HE)染色和免疫组化检测,对比姜黄素治疗前后CIA大鼠炎性细胞、IL-17阳性细胞数量变化.结果 大鼠免疫后2周左右发生CIA.C组大鼠踝关节和趾关节明显肿胀,关节炎评分在实验第21天达到最高分,用姜黄素治疗后关节炎评分逐渐下降,影像学评分明显下降;滑膜细胞增殖减少,组织增厚消失,炎性细胞数和IL-17阳性细胞数量明显少于B组.结论 姜黄素可控制并改善类风湿关节炎症状,降低IL-17表达,对IL-17等炎症性细胞因子异常表达引起的类风湿关节炎有一定的治疗效果.

英文摘要:

Objective To investigate the treatment effect of curcumin on collagen induced arthritis (CIA) in rats and itsmechanism. Methods Fifty five male Wistar rats were randomly assigned to three groupsGroup A was the blank control group,CIA rat mlels were in group B, while CIA rat models with curcumin treatment were in group C. Rats in group B and group Cwere induced by collagen I to establish CIA models. Rats in group C were given curcumin by gastric infusion every two days fromthe 21th day of the cx.ceriment, all the models were observed and the arthritis scores were recorded. X-ray and imaging scores weretaken before and afler the curcumin treatment. All animals were killed at two weeks after treatment, and routine HE andimmunohistochemical staining were taken. Pathological sections and the number of inflammatory cells and interleukin (lL) 17 positive cells were compared with the ones before curcurnin treatment. Results Clinical symptoms could be observed at twoweeks after immunizalion. All ankles and toe joints got swelling while the arthritis score reached the peak in group C and thenreduced gradually after curcumin treatment. The imaging scores became lower after. Synovial cell proliferation andhyperplasia enlargement of the tissue were all reduced and the number of inflammatory ils and IL-17 positive cells weresignificantly decreased. Conclusions The expression of IL-17 and tee symptom of rheumatoid arthritis can be depressed bycurcumin, which will help to cure this disease caused by abnormal expression of IL-17 and other inflammatory cytokines.
