目的对猫脑挫裂伤灶周围神经元在急性期的时间空间规律进行研究。方法采用改良的Feeney法制作猫脑挫裂伤模型,取挫裂伤周皮层组织制备单细胞悬液,Annexin V联合PI标记,应用流式细胞仪分析伤后不同时间挫裂伤周神经元凋亡情况,透射电镜观察细胞超微结构变化。结果脑挫裂伤周围伤后1就有神经元凋亡出现,伤后12h凋亡达高峰,离挫裂伤灶越近,凋亡越重。结论猫脑挫裂伤后周围早期就有凋亡现象,挫裂伤周神经元凋亡的机制和作用仍待进一步研究。
Objective To investigate the temporal and spatial characteristic of neurons around contusion zone in acute period in cats following trauma. Methods Brain contusion model was established as Feeney's model. After operation , Cortical tissues were removed and dissolved . These dissolved cells were retained with Annexin V/PI, and then measured with flow cytometry to analyze neurons apoptosis. The ultrastrure was observed around contusion zone. Results There are apoptotic neurons 1 h after trau- ma, and predominantly expressed at 12 h following injure. Apoptotic neurons were more and more lightly with the distance. Conclusions The present findings confirm the early existence of apoptotic neurons around contusion zone. Further study on the mechanic and function of apoptosis around contusion zone was needed.
