目的探讨低剂量铅对神经细胞粘附分子(NCAM)3个主要亚型(NCAM120、NCAM140、NCAM180)表达的影响。方法用阳离子脂质体瞬时转染法将分别含有NCAM120、NCAM140、NCAM180基因的重组质粒导入到SH-SY5Y细胞中,用1,10μmol/L氯化铅(PbCl2)分别染毒24,48 h,用蛋白印迹(Western blot)法检测3个亚型的表达。结果染毒24,48 h,NCAM120和NCAM180的表达量与阴性对照组比较均降低;随剂量增加,24,48 h NCAM120的灰密度值比从0.580和0.225下降到0.154和0.017,NCAM180从0.086和0.062下降到0.042和0.046,而NCAM140只在10μmol/L中表达下降。结论 PbCl2对NCAM3个亚型的表达有不同程度的影响,其中受铅影响最大的是NCAM120。
Objective To study the impact of lead exposure on the expressions of three main subtypes of neural cell adhesion molecule(NCAM).Methods NCAM120,NCAM140 and NCAM180 recombinant plasmids were transfected into SH-SY5Y cells separately using cationic liposome transient transfection.After identification with immunohistochemistry and Western blot,the cells were co-cultivated with 1μmol/L or 10μmol/L PbCl2 for 24 or 48 hours.Then the expressions of three subtypes of NCAM were detected with Western blot.Results After lead exposure for 24 or 48 hours,the expressions of NCAM120 and NCAM140 were reduced in the cells compared to that of the negative control group.The AO ratios of NCAM120 decreased from 0.580(24hr)and 0.225(48hr)to 0.154 and 0.017;while the AO ratios of NCAM180 decreased from 0.086(24hr)and 0.062(48hr)to 0.042 and 0.046.However,for the cells transfected with NCAM140,only in the group with 10 μmol/L lead exposure,the AO ratios decreased from 1.049(24hr)and 1.027(48hr)to 0.836 and 0.868.Conclusion Lead could influence the expression of the three subtypes of NCAM-NCAM120,NCAM140 and NCAM180,and the expresion of NCAM120 is affected most seriously by lead exposure.
