SUMO是近年发现的类泛素化修饰蛋白,可通过异肽键共价连接到靶蛋白上,影响靶蛋白的细胞内定位、稳定性及与其它蛋白质的相互作用.为研究蛋白质的SUMO化修饰,本文表达并纯化了重组的人SUMO1,制备了兔抗hSUMO1的多克隆抗体.经ELISA和免疫印迹检测,该抗体灵敏度高、特异性好,且能识别内源性SUMO1及其修饰蛋白,可用于SUMO化修饰靶蛋白的鉴定及SUMO化修饰的生物学功能研究.
Small ubiquitin-like modifier (SUMO) is a recent member of ubiquitin-like protein superfamily. As a form of post-translational modification, SUMO attached covalently to target proteins to alter the localization, stability of the target proteins, as well as its interactions with other proteins. To investigate protein SUMOylation, we expressed and purified human SUMO1 , and used to raise high-titer SUMO1-specific polyclonal antiserum in rabbits. The obtained antibodies can recognize endogenous SUMO1 and its association with target protein, thus can be used for the identifications of SUMO target proteins and the studies on biological functions of SUMOylation.
