目的:通过围产期腹腔注射地卓西平马来酸盐(dizocilpine maleate,MK-801)建立精神分裂症(schizophrenia,SZ)发育大鼠模型,探讨不同剂量帕潘立酮(paliperidone,Pal)治疗对SZ发育模型大鼠行为学的影响。方法:将32只出生后7 d(P7 d)的雄性SD仔鼠随机分为对照组、模型组、Pal低剂量组(0.03 mg/kg)、Pal高剂量组(0.3mg/kg),每组8只。模型组、Pal低剂量组、Pal高剂量组仔鼠腹腔注射0.2 mg/kg MK-801,一天1次,连续注射15 d;对照组腹腔注射等体积生理盐水(NS)。P45 d Pal治疗组大鼠腹腔注射0.03mg/kg或0.3 mg/kg Pal溶液,对照组和模型组接受等体积NS注射,一天2次,持续15 d。P60 d测试各组大鼠自发活动、社会交往、前脉冲抑制(pre-pulse inhibition,PPI)和Morris水迷宫等行为学指标。结果:与对照组比较,SZ发育模型大鼠自发活动总路程显著延长(P〈0.01)、社会交往时间显著减少(P〈0.01)、PPI显著降低(P〈0.01)、逃避潜伏期显著延长(P〈0.01)、而在目标象限游泳时间显著缩短(P〈0.01);不同剂量Pal重复处理可以不同程度改善MK-801诱导的行为学异常。结论:围产期MK-801重复处理可引起大鼠成年后自发活动增加、社交功能损害、PPI和空间学习记忆功能缺陷,Pal治疗可以不同程度改善MK-801诱导的上述行为损害。
AIM: To explore the therapeutic effects of different doses of atypical antipsychotic Paliperidone( Pal) on behavioral changes in model rat by establishing the development model of schizophrenia( SZ) by intraperitoneal injection MK-801 in perinatal. METHODS: Thirty-two newborn SD male rats were randomly divided into control group,model group,low-dose Pal group,and high-dose Pal group. SD rats were intraperitoneally injected normal saline( control group) or 0. 2 mg / kg Dizocilpine( MK-801) solution( model group,low-dose Pal group,and high-dose Pal group) at the 7th day after birth( P7),once a day for 15 days. At 45th( P45)day,normal saline( control group and model group)or 0. 03 mg / kg Pal solution( low-dose Pal group) or0. 3 mg / kg Pal solution( high-dose Pal group) were intraperitoneally injected,twice a day for 15 days.Behavior test was conducted from P60 days. RESULTS: Compared with the normal control group,the locomotor activity total distance of SZ developmental model rat was significantly longer( P〈0. 01),social interaction time,pre-pulse inhibition( PPI) and time spend in object quadrant were significantly reduced( P〈0. 01) while the escape latency significant increased( P〈0. 01). Different doses of Pal could improve the MK-801-induced behavioral changes in varying degrees. CONCLUSION: Repeated MK-801 injection in perinatal rats can cause spontaneous activity increase,social interaction dysfunction, PPI damage, spatial learning and memory damage in adult rats,which all can be improved by paliperidone.
