目的探讨骨癌痛大鼠脊髓背角神经元双孔钾离子通道TRESK(TWIK-related spinal cord K+channel,TRESK)表达水平的变化。方法雌性SD大鼠(体重150~180g),随机分为假手术组(Sham组)和骨癌痛组(BCP组)。在术前1d、术后3、5、9、14d分别测量各组大鼠机械缩足阈值(mechanical withdrawal threshold,MWT)的变化,于第14天取腰段脊髓免疫荧光染色及Western blot检测各组大鼠腰段脊髓背角神经元TRESK的表达变化。结果手术前两组大鼠MWT值差异无统计学意义(P〉0.05);BCP组在手术后5dMWT值开始降低,与Sham组相比,差异有统计学意义(P〈0.05)。免疫荧光染色显示TRESK的表达主要位于脊髓背角。Western blot检测结果显示,BCP组的TRESK蛋白表达较Sham组明显减少(P〈0.05)。结论骨癌痛大鼠脊髓背角神经元TRESK表达降低,这种表达的改变可能与骨癌痛发生机制相关。
Objective To investigate the changes in the expression of TWIK-related spinal cord K+ channel(TRESK) in the spinal dorsal horn of rats with bone cancer pain. Methods Adult female Sprague Dawley rats,weighing 150--180 g, were ran- domly divided into two groups:bone cancer pain group(BCP) and sham group (Sham). The mechanical withdrawal threshold (MWT) was measured in all the rats 1 day before and 3,5,9,14 days after surgery by using calibrated Von Frey filaments ap- plied to the incision site. The L4-5 spinal cords of hyperalgesic rats(both obtained at 14 days post surgery) were removed for determination of TRESK expression by immunofluorescence staining and Western blot. Results Allodynia tended to occur at 5 days after surgery,and continued to persist by day 14 post surgery in rats of the BCP group. The immunofluorescence staining showed that TRESK was predominately expressed in the spinal cord dorsal horn. At 14 days after surgery, the expression of TRESK in neurons of the spinal dorsal horn was significantly decreased in BCP group compared to the Sham group. Conclusion The changes in the expression of TRESK in neurons of the spinal dorsal horn may be involved in the development of bone cancer pain.
