目的研究PPAR-1激动剂罗格列酮(ROS)对变应性鼻炎小鼠鼻黏膜半乳糖凝集素-9(galectin-9)和T细胞免疫球蛋白黏蛋白分子-3(tim-3)表达的影响。方法应用卵清蛋白(OVA)致敏建立小鼠变应性鼻炎模型,应用免疫组化及RealTime—PCR方法检NROS对小鼠鼻黏膜中galectin-9和tim-3表达影响,应用ELISA法检测小鼠血清中IL-4、IL-5、IFN-1含量。结果变应性鼻炎(AR)小鼠鼻黏膜中galectin-9和tim-3表达较正常对照组明显增高,罗格列酮及地塞米松治疗后galectin-9和tim-3的表达降低。与正常对照组比较,变应性鼻炎小鼠血清IL-4、IL-5含量明显升高,而IFN-y含量明显减低;罗格列酮组和地塞米松组IL-4和IL-5含量较AR组降低,IFN-y含量上调。结论罗格列酮抑制变应性鼻炎黏膜炎症反应,可能与降低鼻黏膜galectin-9和tim-3的表达相关。。
Objective To research the effect of PPAR- T ligand agonist rosiglitazone on galectin-9 and tim-3 expression in nasal mucosa and contents of ~qammatory factors in serum of allergic rhinitis(AR) mice. Methods Fourty mice were divided averagely into 4 groups: AR group, Rosiglitazone(ROS) group, Dexamethasone(DEX) group and normal group. The mice were immunized by ovalbumin(OVA) at days 1 and 14,, the sensitized mice were exposed to OVA each day from day 21 to 27. The mice of ROS group and DEX group received the test drugs from day 21 to day 27 after the challenge, mice in normal group were exposed to the saline. Immunohistochemical staining was used to examine the galectin-9 and tim-3 expression in nasal mucosa. Real time-PCR was performed to investigate the level of mRNA expression of complement galectin-9 and tim-3 in nasal mucosa. ELISA was performed to investigate the level of proteinum expression of IL-4, IL-5 and IFN- y, in serum. Results Expression levels of galectin-9 and tim-3 were higher in AR group than in N group, Dex and ROS can downregulate their expression. The levels of IL-4 and IL-5 in serum were decreased, but increased for IFN- y in serum in AR group than in N group; however, increased for IL-4 and IL-5, decreased for IFN- y in Dex and ROS groups than in AR group. Conclusion The treated effect of rosiglitazone on allergic rhinitis may be related to inhibition of galectin-9 and tim-3 expression.
