目的:研究复方蜥蜴散不同微粒组合剂(FFXY)治疗对胃癌前病变(PLGC)大鼠凋亡相关蛋白RelA(P65)及IKKβ表达的影响,揭示其治疗PLGC的可能作用机制。方法:将120只SPF级SD大鼠随机分为空白对照组,模型对照组,维酶素治疗组,复方蜥蜴散80目、100目等量混合治疗组,80目治疗组及100目治疗组,每组20只,除空白组外,均采取MNNG配合饥饱失常及情绪刺激综合造模。造模成功后分别作相应处理,应用免疫组化法检测其对胃黏膜组织IKKβ、RelA蛋白表达的影响,并对实验结果进行统计分析。结果:通过图像分析软件对免疫组化图像光密度的测算,1模型组IKKβ、RelA的阳性表达高于空白对照组,有显著性差异(P〈0.01);2其他各治疗组IKKβ、RelA的阳性表达均低于模型对照(P〈0.01);3FFXY各治疗组IKKβ、RelA的阳性表达均低于维酶素治疗组(P〈0.05);480目、100目等量混合组两种因子阳性表达低于80目组和100目组(P〈0.05),80目组和100目组两种因子表达相当。结论:1通过MNNG等综合诱导因素造模成功;2FFXY各组和维酶素组治疗PLGC均有效,其中FFXY各组对PLGC大鼠疗效均优于维酶素组,尤以80目、100目等量混合剂量组疗效最显著。表明FFXY对PLGC胃黏膜损伤可能具有保护、修复和治疗作用,其作用机制可能是通过降低或抑制PLGC组织中IKKβ、RelA的表达,从而促进细胞凋亡。
Objective: To research the effect of different complex lizards powder particle combined agents( FFXY) on PLGC rats apoptosis related proteins RelA( P65) and IKKβ expressions to reveal the mechanism of the treatment of PLGC. Methods: A total of 120 SPF SD rats were randomly divided into blank control group,model control group,vatacoenayme treatment group,complex lizards powder 80 meshes and 100 meshes mixed treatment group,80 meshes treatment group and 100 meshes treatment group,20 in each group,Except the blank group,others all used MNNG,hunger disorders and emotional stimuli comprehensive for building the model. The corresponding processings were adapted after successful model building respectively,using immunohistochemical method to detect the gastric mucosa tissue IKKβ and the influence of the RelA protein expression and the experimental results were analyzed. Results: Through image analysis software to calculate the immunohistochemical image optical density,it showed the model groups' IKKβ and RelA positive expressions were higher than those of the blank control group and there were significant differences( P〈0. 01). The other treatment groups' IKKβ and RelA positive expressions were lower than those of the model control group( P〈0. 01). 3. Each FFXY treatment groups' IKKβ and RelA positive expressions were below that of the vatacoenayme treatment group( P〈0. 05). 80 meshes and 100 meshes mixed group's positive expressions were less than those of the 80 mesh and 100 mesh treatment groups( P〈0. 05). 80 mesh and 100 mesh treatment groups' expressions were the same( P〉0. 05).Conclusion: 1. MNNG and comprehensive induced factors can build the model. 2. FFXY groups and vatacoenayme group were effective for treating PLGC,among which,each FFXY treatment groups' effects were better than that of the vatacoenayme group.
