目的通过观察艾灸预刺激对阿尔茨海默病(AD)模型大鼠细胞周期蛋白CDK2、p21/cip的影响,探讨艾灸预刺激防治AD的作用机制。方法将40只SD大鼠随机分为正常组、假手术组、模型组、预艾灸组,每组各10只,以大鼠海马注入A131—42制作AD模型,预艾灸组采用艾务温和灸“百会”、“肾俞”两穴,免疫组化实验测定大鼠海马区CDK2、p21/cip蛋白的表达。结果预艾灸组经过2个月的预艾灸治疗,CDK2蛋白阳性细胞教相应减少,p21/cip蛋白阳性细胞数相应增加,与模型组比较有显著性差异(P〈0.01)。蛄论艾灸预刺激可调控细胞周期蛋白的表达,通过降低CDK2的表达,增加p21/tip的表达来抑制AD的病理改变,达到防治AD的目的。

英文摘要:

Objective It is to observe the influence of pre-stimulation with moxibustion on cell cycle proteins CDK2 and p21/cip in Alzheimer disease (AD) rat model, and explore the mechanism of pre-moxibustion on prevention and treatment of Alzheimer' s disease. Methods 40 rats were randomly divided into normal group, sham operation group, model group and pre- moxibustion group, there were 10 cases in each group. The AD rat model was established by injecting Aβ1 -42 separately into hippocampus tissue of rats. " Baihui" and "Shenshu" were stimulated in pre- moxibustion group with moxa. The expression of CDK2 and p21/cip in hippocampus tissue of rats were detected by Immunohistochemistry experiments. Results CDK2 pro- tein positive cells were reduced correspondingly and p21/cip protein had a corresponding increase in the number of positive cells by pre-moxibustion treatment. Compared with model group, there was significant differences (P 〈 0.01 ). Conclusion Pre-stimulation of moxibustion can regulate cell cycle-related protein expression, by reducing the expression of CDK2 and in- creasing the expression of p21/cip to inhibit the specific pathological changes of AD, so as to achieve the purpose of combating AD.
