To understand the molecular mechanism of tick blood feeding and pathogen transmission, we cloned a full-length sequence of the immunosuppresant protein P36 gene from Hyalomma asiaticum tick. The cloned gene was 924 bp in length containing an ORF of 708 bp which encodes 235 amino acids. Blast analysis showed the gene shared significant homology with the immunosuppresant protein P36 of Dermacentor andersoni and Amblyomma americanum. The mRNA transcripts of P36 could be detected in the fed tick, but not in unfed tick. The gene was subcloned into expressing vector pGEX-4T-lafter removing the signal peptide and expressed successfully as a GST fusion protein.
