目的 观察三七注射液对阿霉素肾纤维化大鼠肾组织转化生长因子-β(1TGF-β1)和P38 MAPK的影响。方法 120只Sprasue-Dawley大鼠随机分为5组:正常组(=24)、假手术组(=24)、模型组(=24)、三七注射液组(=24)和三七总皂苷组(=24),12周后处死大鼠。免疫组化和Western blot观察大鼠肾组织TGF-β1和P38 MAPK蛋白的表达。结果 正常组和假手术组大鼠肾小管可见极少量的TGF-β1和P38 MAPK阳性蛋白表达;而模型组大鼠肾组织TGF-β1和P38 MAPK阳性蛋白表达上调,与假手术组相比较,差异均有统计学意义(P〈0.01);与模型组相比较,三七注射液组和三七总皂苷组大鼠肾组织,TGF-β1和P38 MAPK蛋白表达明显下调,差异均有统计学意义(P〈0.01)。结论 三七注射液的肾保护作用可能与下调TGF-β1和P38 MAPK水平,抑制TGF-β1/P38 MAPK信号通路活化有关。
[ Objective ] To observe the effect of SanQi injection on the protein expression of Transforming Growth Factor-β1 (TGF-β1), P38 MAPK in the renal tissue of rat model with Adriamycin-induced kidney fibrosis. [Methods] A total of 120 Sprasue-Dawley rats were randomly divided into five groups: normal group (n = 24), sham group (n = 24), model group (n = 24), SanQi injection group (n = 24) and panax notoginseng saponins (PNS) group (n = 24). All the rats were sacrificed after 12 weeks, and the protein expressions of TGF-β1 and P38 MAPK in the renal tissue were detected by immunohistochemistry and Western blot. [Results] The TGF-β1 and P38 MAPK were slightly expressed in the normal group and sham group. However, the protein expressions of TGF-β1 and P38 MAPK in the model group were significantly higher than those in the sham group (P 〈 0.01). The protein expressions of TGF-β1 and P38 MAPK in both SanQi injection group and PNS group were significantly lower than those in the model group(P 〈 0.01). [Conclusions] SanQi injection could provide a nephroprotective role by down-regulating the protein expressions of TGF-β1 and P38 MAPK, and blocking the activation of TGF-β1/P38 MAPK signaling pathway.
