目的:研究曲古抑菌素A(TSA)对胶原诱导性关节炎(CIA)小鼠病情的影响及其机制。方法:将DBA/1雄性小鼠随机分为模型组、TSA组、对照组。免疫34d后对小鼠进行关节炎病变评分、滑膜炎症评分、组织病理检查,通过实时荧光定量PCR及ELISA方法分别检测辅助性T细胞Th1、Th17、Th2细胞的代表性细胞因子干扰素(IFN-γ)、白细胞介素(IL)-17、-4mRNA及蛋白表达水平。结果:TSA组小鼠关节炎指数(2.81±0.10)显著低于模型组(3.63±0.23);TSA组IFN-γ、IL-17 mRNA表达水平(6.39±0.51,2.94±0.40)低于模型组(7.99±0.63,5.87±0.24),IL-4表达水平(2.11±0.15)ng/L高于模型组(1.12±0.07)ng/L。且差异具有统计学意义(P〈0.05)。结论:TSA可能通过抑制Th1和Th17细胞分泌细胞因子IFN-γ及IL-17,增加表达Th2细胞IL-4因子来帮助缓解CIA病情。
Objective:To investigate the effects and mechanism of Trichostatin A(TSA)on the mice model of collagen induced arthritis(CIA).Methods:The DBA/1male mice were randomly divided into model group,TSA group and control group.At the end of 34 days after the injection,the mean arthritic index,the synovitis score and the histopathologic examination were evaluated on the mice.The mRNA and protein expression of IFN-γ,IL-17,and IL-4were detected by realtime PCR and ELISA.Results:The MAI of the TSA group(2.81±0.10)was remarkably lower than that of the model group(3.63±0.23).The mRNA expression of IFN-γ,IL-17 of the TSA group(6.39±0.51,2.94±0.40)were lower than that of the model group(7.99±0.63,5.87±0.24),while IL-4of the TSA group(2.11±0.15)was higher than that of the model group(1.12±0.07),P〈0.05.Conclusion:TSA might mitigate the disease of CIA by inhibiting Th1 and Th17cells secreting cytokine IFN-γand IL-17 as well as increasing Th2 cells secreting cytokine IL-4.
