目的了解趋化因子CXC配体=12(CXCI.12)及其受体CXCR4在子宫内膜癌组织表达及其意义。方法采用RT—PCR方法,检测12例正常子宫内膜组织和29例子宫内膜癌组织CXCI,12、CXCR4mRNA表达,分析子宫内膜癌组织CXCL12、CXCR4mRNA表达与临床病理特征的关系。结果子宫内膜癌组织CXCL12、CXCR4mRNA表达显著高于正常子宫内膜组织(t=3.338、4.174,P〈0.01),且二者表达呈显著正相关(r=0.796,P〈O.01)。子宫内膜癌组织中CXCI。12mRNA表达与FIGO分期、淋巴结转移、细胞分化程度、病理类型及年龄无关(P〉0.05);不同病理分级的子宫内膜癌组织CXCR4mRNA表达差异有统计学意义(F=5.538,P〈O.01),且其表达与子宫内膜癌病理分级呈负相关(r=0.472,P〈0.05);CXCR4tuRNA表达与FIGO分期、淋巴结转移、病理类型及年龄无关(P〉0.05)。结论子宫内膜癌组织中CXCL12、CXCR4mRNA表达上调,提示CXCLt2和CXCR4可能在子宫内膜癌的发生、发展中发挥生物学作用。.
Objective To observe the expressions of ehemokine CXCL12 and its receptor CXCR4 in eridometrial carcino ma (EC) and their significance. Methods The mRNA expression of CXCL12 and CXCR4 in 12 cases of normal endometrial tis- sue and 29 of EC tissue were detected by employing RT-PCR. The relationship between the rnRNA expressions of CXCL12/CXCR4 and clinicopathologie features of EC was analyzed. Results The mRNA expressions of CXCL12 and CXCR4 in EC tissue were markedly higher than that in normal endometrial tissue (t =3. 338,4.174;P 〈0.01), which were positively correlated with each other (r= 0.796,P〈0.01). CXCL16 mRNA expression in EC tissue was not associated with clinical stage, pathologic grade, lymph node metastasis, histological type, and patient's age (P〉0.05). The difference of CXCR4 mRNA expression in different- pathologic-grade EC tissue was significant (F = 5. 535, P 〈 0.01 ), the expression was negatively correlated with the grade (r =0.472,P〈0.05). CXCR4 mRNA expression was not associated with clinical stage, lymph node metastasis, histological type, and patienl's age (P〉0.05). Conclusion In EC tissue, the expressions of CXCL12 and CXCR4 mRNA up-regulate, which prompts that CXCL12 and CXCR4 may play an important role in the development and progression of endometrial carcinoma.
