目的探讨益气活血治法与其中药复方脑络欣通及其拆方(益气方、活血方)对局灶性脑缺血再灌注模型大鼠神经干细胞及相关调节因子的影响,比较其在不同时问位点的作用特点及其机制。方法采用线栓大脑中动脉法制作大鼠局灶性脑缺血再灌注模型,分别观察益气活血、益气、活血三种治法及其代表方药在缺血2h、再灌注1、3、7d的治疗效果;免疫组化染色SABC法检测Nestin、BDNF、EGF蛋白表达。结果(1)Nestin表达情况:缺血再灌注7d时,各治疗组与模型组比较有显著差异;组间比较,仅在缺血再灌注7d时有显著差异。(2)BDNF表达情况:益气活血法组与模型组比较有显著差异(3d、7d),益气法组和活血法组与模型组比较有显著差异(3d或7d);组间比较,缺血再灌注7d时益气活血法组与益气法组和活血法组有显著差异。(3)EGF表达情况:益气活血法组与模型组比较有显著差异(7d);组间比较,7d时益气活血法组与益气法组和活血法组有显著差异。结论经过脑络欣通及其拆方(益气方、活血方)干预后,可使内源性神经干细胞的增殖明显增加,相关调节因子BDNF、EGF的表达也发生不同的变化;脑络欣通在促进神经干细胞的增殖(7d)、促进BDNF(3d、7d)、EGF(7d)表达的效应上优于其拆方(益气方、活血方)。
Objective: To investigate the effects of Naoluoxintong which was representative prescription of supplementing qi and activating blood therapy, the prescription of supplementing qi therapy and the prescription of activating blood therapy on local cerebral ischemia - reperfusion injury rats. To observe the changes of neural stem cells (NSCs) and correlated regulatory factors, and to compare the effects and mechanism of Naoluoxintong and other prescriptions on different time points. Methods: Models of local cerebral ischemiareperfusion were established by threa doccluding the MCA. The time points were observed at 1, 3 and 7d after reperfusion following 2h of MCAO respectively. The expression levels of Nestin, BDNF, EGF were measured by immunohistochemical staining. Results: ( 1 )The expression level of Nestin: the expression of Nestin of treatment groups increased significantly compared with model group at 7d, and Yiqihuoxue treatment group increased which has quiet variability compared with Yiqi treatment group and Huoxue treatment group. (2) The expression level of BDNF: the expression of BDNF of Yiqihuoxue treatment group has quiet variability compared with model group at 3d and 7d, Yiqi treatment group and Huoxue treatment group also have quiet variability compared with model group at 3d or 7d. Yiqihuoxue treatment group increased significantly which has quiet variability compared with Yiqi treatment group and Huoxue treatment group at 7d. (3)The expression level of EGF: The express of EGF of Yiqihuoxue treatment group has quiet variability compared with model group at 7d, and Yiqihuoxue treatment group also has quiet variability compared with Yiqi treatment group and Huoxue treatment group at 7d.
