目的探讨核因子-κBB(NF-κB)和肿瘤坏死因子-α(TNF-α)在小鼠抗烟曲霉菌早期感染中的作用。方法将小鼠分4组:正常组、免疫抑制组、正常+烟曲霉菌接种组和IPA模型组。小鼠鼻吸人烟曲霉菌48h处死,取肺组织用western blot法检测胞核NF-κB p65蛋白和细胞中TNF-d的表达。结果正常+烟曲霉菌接种组和IPA模型组小鼠肺组织细胞核NF-κB p65蛋白和细胞中TNF—α表达的量高于正常组和免疫抑制组;正常组小鼠中检出NF-κB p65蛋白和TNF-α表达的量高于免疫抑制组;免疫抑制组肺组织细胞核NF-κB p65蛋白量最低,并且未检出TNF-α的表达。结论烟曲霉菌早期感染可激活小鼠肺组织中NF-κB并上调TNF-α蛋白的表达。
Objective To explore the role of NF-kB and TNF-α in controlling early infection of aspergillus fumigatus in mice. Methods The mice were divided into four groups,including group of normal mice,group of immunosuppresive mice, group of normal mice infected with A. fumigatus and group of IPA mice. The mice were sacrificed at 48 h after infection to collect the lung tissue for detection of nucleus NF-kB p65 and expression of TNF-α with western blot. Results The level of NF-kB p65 and TNF-α expression in the group of normal mice infected with A. fumigatus and the group of IPA mice were higher than those in the group of normal mice and the group of immunosuppresive mice in western blot. The physiological level of NF-kB p65 and expression of TNF-α in the group of normal mice were detected and higher than in the group of immunosuppresive mice. The expression of TNF-α was not detectable via western blot in the group of immunosuppresive mice. Conclusion The early infection of aspergillus fumigatus could activate NF-kB and upregulate TNF-α expression in the lungs of experimental mice.
